In Histopathology, Xylene is a clearing agent used in tissue processing which is toxic and carginogenic thereby posing a risk to the health of medical workers. Hence, A known non-toxic substitute for xylene is  d-limonene. D-Limonene is the major constituent of citrus peel oils such as calamansi. This research aimed to determine the clearing property of Calamansi (Citrus japonica) peel extract in histotechniques. The d-limonene was extracted from the peel of calamansi by the process of steam distillation at the Chemical and Energy Division of the Department of Science and Technology. The prepared extract was used as a clearing agent in tissue processing. The tissue samples were graded based on transparency, hardness and staining. Out of 10 tissue samples, a computed p-value of 0.003 (based on tissue transparency), 0.533 (based on tissue hardness) and 0.418 (base on tissue staining) was acquired. The result of the t-test indicated a significant difference between the calamansi extract and xylene (control) base on tissue  transparency but no significant difference base on tissue hardness and tissue staining.
